Keeping pace with the increasing demand for high quality drug candidates in pharmaceutical research: Development of a new two-step preparative tandem high performance chromatographic system for the purification of antibodies.
In the field of biological drug development, high throughput (HT) technology is the key to identifying the most promising therapeutic candidates in an efficient manner and time-competitive market. While efficient cloning and expression of the existing methods, HT downstream processing mainly relies on applying liquid handling workstation miniature chromatography column or resin-based 96-well plate to shorten the development time. However, there is still a shortage of generic, preparative chromatographic method without aggregates and endotoxin with sufficient throughput.
The only thing that really generic antibody purification strategies including the removal of efficient dimer consisting of Protein A capture followed by size exclusion chromatography (SEC) as a polishing step. Other polishing methods, including IEX, HIC, and CHT, requires fine tuning specific antibodies. However, the standard setup preparative SEC tends to be somewhat time consuming, and limits throughput. In this work, we designed a unique chromatography setup enables a two-step purification of IgG unattended on a milligram scale directly from the 35 mL cell supernatant clarified, processing up to 48 samples in 44.0 hours.
By introducing the silica-based SEC columns, a preparative SEC can be accelerated. By further developing the HT two-step preparative Protein A / alternating column regeneration SEC system using the Agilent 1260 Infinity LC components, mAbs may be purified generally by two chromatographic steps in 55 minutes. In this way, by using a valve 2-position / 10-port and two quaternary pump, two SEC column can be run in parallel, excluding cleaning and balancing the phase of the actual cycle time. With further implement the third pump, Protein step can be run independently, resulting in a time-optimized nesting process.
By introducing CETAC ASX-520 autosampler, 48 samples can be run automatically without user intervention more than two business days. Advanced highly reproducible system for all human IgG1 antibodies tested, easily produce scale milligrams enough material for a full characterization of the antibodies and for their use in in vitro and in vivo activity assessment.
